Role of endothelin-1 in clinical hypertension: 20 years on. Hypertension. 2008;52 :452-9. 4. Luscher TF, Barton M. Endothelins and endothelin receptor antagonists

Endothelin-1 (ET1) is a 21–amino acid peptide secreted by vascular endothelial cells in response to stimuli, including pulsatile stretch, sheer stress, neurohormones, cytokines, growth factors, and thrombin. The secretion of ET1 has been shown to be increased by hypoxemia in humans. Endothelin-1 (ET-1) is a 21 amino acid peptide with diverse biological activity that has been implicated in numerous diseases. ET-1 is a potent mitogen regulator of smooth muscle tone, and inflammatory mediator that may play a key role in diseases of the airways, pulmonary circulation, and inflammatory lung diseases, both acute and chronic. At the same time, endogenous endothelin-1 acts through endothelial ET(B) receptors to stimulate formation of nitric oxide tonically and to oppose vasoconstriction. In view of the multiple cardiovascular actions of endothelin-1, there has been much interest in its contribution to the pathophysiology of hypertension. Rationale: Endothelin-1 (ET-1) is increased in patients with high-altitude pulmonary edema and acute respiratory distress syndrome, and these patients have decreased alveolar fluid reabsorption (AFR).

Changa, Endothelin is a potent vasoactive peptide occurring in three isotypes, ET-1, ET-2, and ET-3. Through its two main receptors, endothelin A and endothelin B, it is responsible for a variety of physiological functions, primarily blood flow control. Recent evidence from both human and animal models shows involvement of endothelin in diabetes, retinal circulation, and optic neuropathies. Endothelin 1 is one of three isoforms of human endothelin. Endothelins are peptides with receptors and effects in many body organs. Endothelin constricts blood vessels and raises blood pressure.

Generation of Hypo/Hypermorphic Mice for Endothelin-1. As a first step in generating modified forms of the Edn1 gene expressing low or high levels of endothelin-1 for use in studies of the effects of both loss of function and gain of function of endothelin-1, we first compared the relative effects on mRNA stability of the 3′ UTR of WT gene (Edn1) with a panel of other 3′ UTRs, as described

Elevated levels of the vasocontrictor peptide endothelin-1 have been demonstrated in various pathological conditions that are characterized by sodium retention and/or renal vasoconstriction, such as heart failure, hepatorenal syndrome, renal Endothelin-1 functions as an endogenous chemical affecting vascular tone across the organs in the body. All three endothelins are classified as having vascular homeostasis functions. They are involved in cardiovascular function, electrolyte homeostasis, and many other mechanisms on a cellular level. ET1 is released constitutively from endothelial cells and regulates vascular function. These three isopeptides mediate their biological functions by ET α and ET β receptor subtypes [ 169, 170 ]. The ET α is located on vascular smooth muscle cells and ET β on vascular endothelial cells. The receptors function in opposition.

Brain capillary endothelial cells (BCEC), the principal components of the blood—brain barrier (BBB) are interconnected by tight junctions, possess polar membranes and a paucity of pinocytotic vesicles in contrast to capillaries in peripheral organs. 1 BCEC’s unique features embrace specialized transport and carrier systems as well as enzymes associated with various metabolic The Relationship between Circulating Biomarkers of Nitric Oxide and Endothelin-1 and Hemodynamic Function in Obstructive Sleep Apnea by Brian John Hawkins Committee Chair: William G.
